The purpose of the present invention is to provide a manufacturing method of a protein supermolecular assembly, which can effectively obtain a macromolecular protein assembly. The present invention relates to a manufacturing method of a supermolecular assembly; and to a method for displaying protein on a supermolecular assembly, and the manufacturing method of a supermolecular assembly comprises the following step: (a) manufacturing plural assembly monomers which comprise (i) a first split fragment including strands 1-10 of green fluorescent protein (GFP), and (ii) a second split fragment including a strand 11 of GFP, coupled to the end of the first split fragment; and (b) performing a self-assembly process of the plural assembly monomers to manufacture a supermolecular assembly. Furthermore, the present invention relates to a method for refining a polydisperse supermolecular assembly into a monodisperse supermolecular assembly, which comprises the following steps: (a) reducing the net charge of an assembly monomer to mutate the assembly monomer so that the value of a negative charge is increased; (b) performing a self-assembly process of plural assembly monomers to manufacture a polydisperse supermolecular assembly which is negatively charged; (c) separating the polydisperse supermolecular assembly according to the size of the assembly; and (d) recovering a monodisperse supermolecular assembly.
