Disclosed is an electrochemical biosensor based on a nucleic acid aptamer / nanosilver probe and an EXO I enzyme. A silver nanoparticle modified with a nucleic acid aptamer that has both a function of recognizing a target substance and a function of generating an electrochemical signal is used as a bioprobe to detect a target biomolecule. A complementary probe and an EXO I enzyme are started by the target substance. Using the complementary pairing principle of DNA, with the aid of cyclic cleavage amplification by, the probe collects on the surface of the gold electrode and the higher the concentration of the target biomolecule, the more the nucleic acid aptamer / nanosilver probe agglutinates. Furthermore, by introducing the target cycle amplification mechanism of EXO I exonuclease into the recognition process of the target substance, the electrochemical biosensor can detect the target biological substance with high sensitivity and efficiency.
