The present invention discloses an electrochemical biosensor based on a nucleic acid aptamer / nanometer silver probe and an EXO-I enzyme which comprises using nanometer silver particles modified with a nucleic acid aptamer and detecting both a target substance and a nucleic acid aptamer also capable of generating an electrochemical signal, as a biosensor for detection of a Zielbiomoleküls when triggered by a target substance with the support of a cyclic cleavage and amplification using a complementary probe and an EXO-I enzyme by means of a complementary DNA mating principle, an accumulation of Probe is provided on the surface of a gold electrode, wherein at a greater concentration of the target biomolecule, a higher collection density of the nucleic acid aptamer / nanometer silver probe is effected, at the same time in the detection of the target substance, a cyclic amplification of the target by means of ei ner EXO-I exonuclease is introduced to allow a sensitive and efficient detection of the target biomaterial by such an electrochemical sensor.
