A one pot cascade cyclization process for synthesis of 4-substituted quinoline pharmacophore analogues (Formula 1) such as natural drug molecule Graveoline; said process of synthesizing functionalized quinolines comprises a metal-free, Lewis/Bronsted acid mediated segment coupling of alcohol, o-azido aldehyde along with alkynes. This process involves a sequence of oxonium ion formation (Int-A), C-C bond formation through its trapping by alkyne (Int-B), followed by C-N bond formation with concomitant extrusion of nitrogen (Int-C). This metal-free method has led to new procedure for the synthesis of various 4-substituted quinoline pharmacophore analogues, drug molecules and natural products. ¦¦¦¦¦¦¦¦ Formula (1)
