The present invention relates to a medium composition for culturing regulated T cells derived from peripheral blood mononuclear cells, and a method for culturing regulated T cells by using the same. More specifically, it has been confirmed that the differentiation of regulated T cells is promoted and the mass-proliferation is possible when an anti-CD3 monoclonal antibody, an anti-CD28 monoclonal antibody, TGF-β, and afzelin are included in the medium composition. In addition, it has been confirmed that regulated T cells proliferated by the method of the present invention can significantly inhibit the activity and proliferation of target immune cells. To this end, the present invention provides a medium composition based on an afzelin and a method for culturing regulated T cells by using the same. In addition, regulated T cells derived from peripheral blood mononuclear cells, which are cultured by the method according to the present invention, can be used for a pharmaceutical composition which is very effective to treat autoimmune diseases. Moreover, the medium composition of the present invention can autonomously treat autoimmune diseases.
