Provided are a biomarker MEL-18 for Human epidermal growth factor receptor2 (HER2) positive cancer and anti-HER2 therapies, and a use thereof. According to the present invention, the MEL-18 is a prognostic and diagnostic factor for the response in an individual to an anti-HER2-targeted therapeutic agent in HER2 positive cancer and can be used in conjunction with the HER2-targeted therapeutic agent in the individual with HER2 positive cancer. Therefore, by determining co-administration with the HER2-targeted therapeutic agent or injection of ADAM 10/17 inhibitors, it is possible to improve therapeutic effects and overcome resistance to the HER2 therapeutic agents, thereby effectively treating HER2 positive cancer.
