The present invention discloses a heat-resistant protective agent, comprising, in mass percentage, 1-5% of raffinose, 5-10% of maltose, 15-30% of sucrose, 1-5% of lactose, 1-5% of glucose, 0.1-1.5% of polysorbate 80, 0.1-0.5% of polyethylene glycol 8000, 0.5-3% of tyrosine, and 3-6% of silk fibroin, the balance being water for injection. The present invention further discloses a preparation method for the heat-resistant protective agent. The present invention still further discloses a live swine fever vaccine allowing for preservation at room temperature and a preparation method therefor, comprising mixing the heat-resistant protective agent and a live swine fever virus solution, followed by gradient vacuum drying to give a live swine fever vaccine. The vaccine prepared by the present invention has an appearance of a dense dry foam shape, and exhibits a glass-like layered structure under a scanning electron microscope, and has a glass transition temperature of up to 50°C or higher, 30°C higher than that of conventional freeze-dried vaccines, and can be stored for 3 months at 37°C, or for one year at 25°C.
