2 RPLC-SFC SYSTEMS AND METHODS FOR TWO-DIMENSIONAL RPLC-SFC CHROMATOGRAPHY

Two-dimensional chromatography systems and methods are provided for separating and / or analyzing complex mixtures of organic compounds. In particular, a two-dimensional reversed phase liquid chromatography (RPLC) -supercritical fluid chromatography (SFC) system including a trapping column is described at an interface that collects eluted analytes from a first dimensional chromatograph while allowing the RPLC mobile phase to pass through. Interest peaks from the RPLC dimensional column are effectively focused as abrupt concentration pulses in the trapping column, which is subsequently injected into the second dimensional SFC column. The system can be used for simultaneous archival and chiral analysis of pharmaceutical compounds. The first dimensional RPLC separation provides an achiral purity result and the second dimensional SFC separation provides a chiral purity result (enantiomeric excess).
