The present invention relates to a method of screening a disease treating candidate material by using a combination of calcium and phosphatidylinositol phosphate. A method of screening a metabolic disease treating candidate material of the present invention comprises: a step 1) of treating cells having an increased calcium concentration therein with a test substance; a step 2) of checking whether calcium and phosphatidylinositol phosphate (PIP) have been combined or not in the calcium concentration-increased cells of the step 1); and a step 3) of sorting the test substance which inhibits a combination of calcium and PIP in the step 2). Specifically, it has been confirmed in the present invention that concentration of calcium is increased in an insulin resistance animal model induced by obesity, and the increased calcium concentration inhibits a movement of Akt protein including a PH domain to a cell membrane and a signal transduction mechanism of the Akt protein, while protein including a C2 domain in the animal model is moved to the cell membrane by being combined with a complex of calcium and PIP under a high concentration of calcium. Therefore, confirming whether calcium and PIP have been combined or not may be useful in screening a candidate substance for treating metabolic disease, cancer or hypertension.
