The present invention relates to a Schizochytrium sp. ABC-101 strain, a microalgae producing bio-oil containing a high concentration of DHA, and a method of producing DHA using the strain. In the present invention, when cultured in specific medium conditions, Schizochytrium sp. ABC-101 having a high yield of bio-oil with high DHA content has been newly isolated. The novel Schizochytrium sp. ABC-101 strain of the present invention is very useful in related industries by making a significant contribution to the production of biomass and the production process of useful substances using the same as a raw material.
