The present invention relates to a real-time PCR method and apparatus. More specifically, the present invention relates to a real-time PCR method capable of performing multi-detection with a single wavelength, and a real-time PCR apparatus therefor. To this end, a probe-type fluorescent material and an intercalator-type fluorescent material which emit fluorescence in the same wavelength band are mixed to be used in response to excitation light emitted from a single wavelength light source so as to enable multi-detection with a single wavelength. The fluorescence emitted from the probe-type fluorescent material is measured in real time to quantify an amplified product of a target gene in a DNA denaturing step. In a melting curve measuring step, the detection of the target gene or an internal control gene is confirmed by using a melting temperature (Tm) of a melting curve showing that the fluorescence emitted from the intercalator-type fluorescent material decreases according to an increase in the temperature, and the reliability of a PCR process and the apparatus may be verified.
