The present invention provides a rifeteglast preparation useful for treating an immune-related disease of the ocular surface. The formulations and methods provided herein are particularly useful for treating anterior segment tissue. The present invention particularly provides a method of treating an immune-related disease of the ocular surface in a subject. In some embodiments, the methods of the present invention are effective in formulations that provide a maximal concentration of rifitegrast (Cmax) of greater than about 5190 ng / mL in anterior segment tissue of the eye for a 1.75 mg dose of rifitegrast. It may include topically administering an amount of rifitegrast or a pharmaceutically acceptable salt thereof to the subject's eye.
