Methods of developing a protein expression profile in a biological sample offormalinfixed tumor tissue obtained from a subject are provided herein. SRM/MRM assaysprovidedherein can be used to detect and quantitate proteins involved in cellularprocesses including celldivision, cellular differentiation, cell growth inhibition, cellularmetabolism, cell signaling, andtumor immune response/modulation directly in a subject's tumor tissue. TheSRM/MRM assayscan provide a tumor tissue profile of the entire tissue microenvironment,regardless of cellularorigin of expression, informing an optimal cancer therapy treatment for thesubject.
