The present invention relates to a method for manufacturing a specimen having a uniform thickness of a homogeneous water-insoluble substance sample and a method for quantitatively analyzing a water-insoluble substance by performing MALDI mass spectrometry on the specimen. Specifically, the specimen may be manufactured by adding a volatile solvent to a mixture of the water-insoluble substance and a matrix, placing the sample, which is obtained by dispersing the mixture of the water-insoluble substance and the matrix until the solvent is volatilized, in a pellet cup made of a water-soluble substance, pressing the sample with the same pressure, and then dissolving the pellet cup with water.
