Toxicological Evaluation of the Weekly Oral Administration of 2,2'-(9-10-Anthrylenedimethylene) -Bis-2-Pseudothiourea Dihydrochloride Dihydrate (Nsc-56054)

NSC-56054 10 mg/kg/day (1.25 mg/ml in 25% propylene glycol) divided in 3 daily doses, was administered to one dog, once a week for 11 weeks. Another dog was treated with the control vehicle only. At the end of the treatment period, both dogs developed a slight anemia, an effect which appears, then, to have been produced from propylene glycol, which was part of the vehicle. The dog which was treated with NSC-56054 vomited after drug administration, and temporarily lost weight. The clinical chemical investigation of the blood failed to reveal significant changes. The histopathological evaluation showed hydropic changes of the liver with small collections of lymphocytes, observations which indicate minimal liver pathology. All these observations lead to the conclusion that NSC-56054 at the investigated regimen failed to produce serious toxic effects, whereas propylene glycol produced slight anemia. (Author)
