Toxicity Studies on Nsc 63878, Cytosine, 1-Beta-D-Arabinofuranosyl-, Hydrochloride, Formulated in Saline, Propylene Glycol, Acid Mantle Creme, Aquaphoraqua and Fifty Per Cent Ethanol Following Topical Application to the Intact and Abraded Skin of Rats, Mice and Guinea Pigs

Compound NSC 63873, Cytosine, 1-beta-D-arabinofuranosyl-, hydrochloride, was applied topically to the intact and abraded skin of mice, rats and guinea pigs in a dose of 60 mg/kg/day for five days using vehicles of Physiological Saline, Propylene Glycol, Acid Mantle Creme, Aquaphoraqua (Aquaphor) and fifty per cent Ethanol. These dose regimens did not produce any overt toxic symptoms in the animals nor any significant alterations in total leucocyte counts except for a decrease in total leucocyte count noted in (1) the abraded skin guinea pigs treated with the drug in propylene glycol, (2) the intact skin mice treated with the drug in aquaphoraqua and (3) the intact skin mice treated with the drug in fifty per cent ethanol. These WBC changes were reversible at the end of the twenty-eight day observation period except for the mice referred to above in (2), but the WBC counts of these animals noted at the end of the treatment and observation periods were well within normal limits for this species. In none of the animals treated with the drug in the various vehicles was there any demonstrable gross or histologic pathology noted in the bone marrow, spleen, gastrointestinal mucosa, or sites of application.
