NSC 121 210 Dehydroepiandrosterone - Mustard, (DHEA Mustard): Preclinical Pharmacological Evaluation of 14Oral,Daily Consecutive Treatments in Rhesus Monkeys.

Androst-5-en-17-one, 3beta-hydroxy-,(p-(bis(2-chloroethyl) amino) phenyl) acetate,NSC-121 210 = DHEA mustard, a compound with antitumor activity was submitted to a preclinical pharmacological evaluation in 26Rhesus monkeys at daily doses of 2.35to 300mg/kg in sesame oil 1.5ml/kg. Treatment produced reversible lymphopenia and hypocellularity of the lymphoid tissues (24/26monkeys),less frequent bone marrow toxicity and occasional renal,hepatic and intestinal toxicity. However 4monkeys treated at various dose levels died from renal or intestinal lesions which in part resulted from a possible activation of a latent Shigellosis. Observation of non dose related toxicity in monkeys agrees with the toxicity of DHEA mustard in dogs.
