Cooperative Study of Spontaneously-Occurring Factor VIII Inhibitors in Patients with Hemophilia A

This contract was part of a multicenter effort aimed at defining the natural history of inhibitors developing in hemophiliacs. A total of 1511 patients were entered into the study, and the final data analysis will be reported from the contract for the reference laboratory and coordinating center, contract number. In addition, a controlled, randomized, double blind study was carried out to determine the efficacy of prothrombin complex concentrates (PCC) in treatment of joint hemorrhage in hemophiliacs with inhibitors. The conclusion of this study was that while these concentrates are only partially effective, their continued use in this setting is justified in the absence of any other effective and readily available form of therapy. The University of North Carolina entered 181 patients into the overall study and 44 treatment episodes into the trial of PCC and also tested patients for liver function and the presence of hepatitis B.
