Task 1 Preclinical Single and Five Daily Dose, Range-Finding and Lethality Studies in CD2F1 Mice, and Toxicity Studies in Mice and Beagle Dogs Administered NSC-305884

Single dose range-finding and lethality studies based upon intravenous administration of NSC-305884 to CD2Fl mice are reported. Combined sex LD10, LD50 and LD90 values produced were 80.37, 96.55 and 115.99 mg/kg (241.11, 289.65, and 347.97 mg/sq.m.) respectively. Toxic signs including exophthalmus, loss of bladder control, irregular breathing, tremors, clonic/tonic convulsions, gasping, and lethargy were immediate in onset, lasted for no more than a few hours, and if not lethal, were readily and completely reversible. Five daily dose range-finding and lethality studies are also reported.
