The goal of the research is the development and laboratory demonstration of reagents and technology for the surface modification of commercially developed substitute anatomical materials to provide them with the proclivity for forming stable natural surface lining after implantation or in cell culture before implantation, through rapid spontaneous overgrowth with endothelial cells. The Phase I objectives were to take commercial substitute blood vessels with optimized mechanical properties and modify their intimal surface without damage to the physical properties, in order to promote endothelial cell migration, attachment, and stable growth. Phase I results show that plastic surfaces treated with human plasma fibronectin significantly enhanced the attachment, growth, and spreading of bovine endothelial cells from fetal heart and cornea. Preliminary data indicate superior storage stability for the photocoupled vs. adsorbed fibronectin on wet surfaces.
