The synthesis and characterization of the poly (alpha-amino acids) is discussed. An improved esterification method is presented for the preparation of L-glutamic acid and L-aspartic acid omega-esters, which are the starting compounds in the synthesis of the amino acid (co)-polymers. The synthesis of high molecular weight poly(alpha-L-glutamic acid), using either new poly(gamma-benzyl-L-glutamate) debenzylation methods or novel L-glutamic acid side-chain protecting groups, is described. Also described is the synthesis and characterization of homo- and copolymers of gamma-piperonyl-L-glutamate and gamma-ethyl-L-glutamate. Subjects discussed include the effect of the polymerization conditions (type of solvent, type and concentration of initiator) on the (co)-polymer molecular weight, the monomer reactivity ratios, and the (co)-polymer molecular weight, the monomer reactivity ratios, and the (co)polymerization kinetics. Next, the synthesis and characterization of the poly(omega-hydroxy acid) poly(L-lactide) is described.
