Depletion of Neuroblastoma Cells from Bone Marrow with Monoclonal Antibodies and Magnetic Immunobeads

In this pre-clinical study, we have investigated removing tumor cells from bone marrow with monoclonal antibodies and magnetic immunobeads (Molday, et al 1977; Rembaum, et al 1982; Treleaven, et al 1984). The following parameters were studied: (1) labelling neuroblastoma cell surfaces with monoclonal antibodies; (2) coating magnetizable beads with goat anti-mouse immunoglobulin; (3) forming bead-tumor cell conjugates; (4) removing bead-tumor cell conjugates with cobalt magnets; and (5) recovery of normal marrow cells. We demonstrated that one thousand to ten thousand-fold depletion of neuroblastoma cells can be reproducibly achieved with excellent recovery of myeloid stem cells (CFU-G,M) and total mononuclear cells.
