Cobalt (III) chelates were investigated as potential reactivators for GD-inhibited aged acetylcholinesterase. Four cobalt (III) amine chelate complexes were synthesized, characterized, and submitted to WRAIR for in vivo testing. The compounds are active catalysts for the hydrolysis of anionic dealkylated phosphonate esters, which are models for GD-inhibited aged enzyme. However, the compounds are not active catalysts for the hydrolysis of neutral phosphonate esters, which are models for GD-inhibited unaged enzymes. Thus, these cobalt (III) chelates would be effective reactivators only in combination with more traditional therapies. A quadrupole pH stat was designed, constructed, and interfaced with a computer system to assist in the rapid acquisition of accurate kinetic data.
