SCREENING AND OPTIMISATION OF FERMENTATION MEDIA OF Marasmius androsaceus USING RESPONSE SURFACE METHODOLOGY

Marasmius androsaceus, a traditional Chinese medicine, has applications in tendon relaxation, pain alleviation and antihypertension in China for thousand years. However, its large-scale application both as a medicine and a food is limited by immature artificial cultivation technology. The optimum fermentation medium of Marasmius androsaceus was not reported yet. In the present study, by using desirability function, four indices (mycelium yield, the contents of intracellular adenosine, mannitol and polysaccharide) were united into one response value (Da). Based on the changes of Da value, three parameters (sucrose, yeast extract powder and KH2PO4) selected via the Plackett-Burman (PB) design were further optimised by response surface methodology (RSM). The optimum medium for Marasmius androsaceus submerge fermentation consisted of: 24.1 g/l sucrose, 10 g/l peptone, 11.3 g/l yeast extract powder, 1 g/l MgSO4 center dot 7H(2)O, 0.955 g/l KH2PO4 center dot 3H(2)O, and 0.1 g/l vitamin B-1. The average optimum Da value was 0.4058 +/- 0.0025. Our finding provides experimental evidence that PB design and RSM are effective tools for mathematical modelling and factor analysis of medium optimisation process.
