Synthesis of 3-(1-Adamantyl)-4-methoxyphenylboric Acid

3-(1-Adamantyl)-4-methoxyphenylboric acid is the key intermediate in the synthesis of active pharmaceutical ingredient (API) 6-[3-(1-adamantyl)-4-meth-oxyphenyl]-2-naphthoic acid (Adapalene). We found that a direct Grignard synthesis involving 2-(1-adamantyl)-4-bromoanisole in the presence of dry lithium chloride led to increase in the 3-(1-adamantyl)-4-methoxyphenylboric acid yield to 70-72%.
