摘要

Fluorescence microscopy is among the most widely used imaging methods in biology. However, conventional light microscopy cannot resolve many structures inside cells because its horizontal resolution is limited to approximately 200 nm. 3D STORM is a recently developed super-resolution technique that can achieve 20-30 nm lateral resolution and 50-60 nm axial resolution, enabling elucidation of previously unresolvable sub-cellular structure. Expansion to multiple fluorophores provides the specificity to interrogate individual biomolecular interactions and to test previous assumptions of co-localization.
