The validation of the proposed health claims of a probiotic strain is best done by an amalgamation of probiogenomics to predict the functional capacities based on genome annotation and experimental clinical evidence. The aim was to predict the functionality of Lactobacillus helveticus MTCC 5463 for vitamins (biotin and folic acid) and exopolysaccharide (EPS) production. The percentage adhesion of MTCC 5463 to HT29 cells was 63% with EPS production of 193 mg/l. The genome carried adhesion causal genessuch as those for putative sortase, aggregation promoting, fibronectin binding, S-layer, mucus binding proteins, glycosyltransferases, lipopolysaccharides, capsular and EPS biosynthesis proteins. Folic acid production estimated by HPLC was 123.0 u.g/1,while the amount of biotin determined using ELISA was 0.23 u.g/g fermented milk. The vitamin biosynthetic cluster included dihydrofolate reductase, 5,10-methylene-tetrahydrofolate cyclohydrolase, fumarate reductase, cobyric acid, thiamine biosynthesis protein and dihydrofolate reductase. Identification of genetic determinants of functionality can augment future bioengineering of the strain.
