摘要

The aim of the present study was to produce live kids from in vitro fertilized goat oocytes after transfer to recipients using laparoscopy. Total 5212 oocytes were collected from slaughtered goat ovaries and 2266 A & B grade oocytes matured in mediumcontaining TCM- 799 plus 5 ug/ml FSH, WO ug/ml LH, 1 ug/ml estradiol-17 beta, 10% inactivated estrus goat serum and 3% BSA in 5% CO_2 in air at 38.5°C with maximum humidity for 27 h. In-vitro fertilization was performed with 27 h matured oocytes and capacitated spermatozoa. Total 494 cleaved embryos developed and same cultured in embryo development medium containing TCM 199 plus 100 mg/ml L- Glutamine, 27 mg/ml sodium pyruvate, 50 mg/ml gentamycin, 3 mg/ml fat free BSA and 10% FCS with goat oviductal epithelial cells. Recipient goats were synchronized with two i.m. injections of PGF_2a (Dose 15 mg/goat; Lutalyse~(R)) 10 days apart. In the present study, 494 two cells to morula stage embryos were produced and 80 eight cells to morula stage embryos weretransferred puncturing uterotubal junction of 14 recipient goats using laparoscope. Four normal kids were born after completion of pregnancy. One recipient delivered twin kids whereas other two recipients delivered single kid. From the present study; itcan be concluded that normal kids were born in Indian breed of goat from IVF embryos after transfer to synchronize recipient goats using laparoscopy.
