摘要

Salmonella infection caused by Salmonella enterica serovar Typhimurium is one of the causes of food-borne disease in humans. An effective way to reduce food poisoning due to Salmonella would be to breed chickens more resistant to Salmonella. In the present study, we studied the genetic resistance of chickens selected for PHA-P response against Salmonella enterica serovar Typhimurium. Chicken o'rthologues of the T helper 1 (Th 1) cytokines interferon-y (IFN-y) and interleukin-2 (IL-2), Th2 cytokine, IL-10, the pro-inflammatory cytokine IL-6 were quantified in peripheral blood mononuclear cells (PBMC) from chicken lines divergent to DTH response (HCMI and LCMI lines) post Salmonella challenge, by real time PCR. The expression of IL-6 and IFN-y was significantly higher in HCMI line as compared to LCMI line. However, significantly higher levels of IL-2 and IL-10 expression were noticed in LCMI line when compared to HCMI line. The higher level expression of pro-inflammatory cytokines in HCMI line suggest the genetic resistance against Salmonella. The down regulation of IL-2 in PBMC of HC"MI line might be due to the higher levels of IFN-y. Further, the higher levels of IL-10 expression might be a reason for the low expression of pro-inflammatory cytokines in LCMI line. These findings suggest that the selection based on PHA-P response modulated the cytokine responses in PBMCs of chicken lines. The selection for in-vivo PHA-P response might be exploited for enhancing immunocompetent of the flock againstSalmonella.
