Simultaneous Determination of Saponins and Anthraquinones in the Herb-Pair of Ginseng and Ploygonum Multiforum by HPLC-DAD

An HPLC-DAD method for simultaneous determination of ginsenosides and anthraquinone derivatives was developed and validated for the investigation of the compatibility of Ginseng and Radix Polygoni Multiflori Preparata (RPMP) herb-pair. Ten extracts of Ginseng-RPMP herb-pair samples with different ratios obtained by ultrasonic extraction and percolation were investigated. The quantitation of ginsenoside Re, ginsenoside Rg1, ginsenoside Rb1, rhein, emodin and physcion in the extracts from Ginseng-RPMP herb-pair were carried out with acceptable multiple correlation coefficients (R~2 ≥ 0.96) and low limits of detection (ranged from 0.085 μg to 0.520 μg). The mean recoveries of the components were 87.9% to 106.7%. Higher extracting efficiency and better repeatability were obtained by percolation extraction than those by ultrasonic extraction. The results showed that the amounts of the bioactive components in the herb-pair were not simple addition of those in Ginseng and RPMP herbs. Some interaction may exist among these constituents. To elucidate the mechanism of action, more data and deep investigation need. The results obtained in the present work can provide useful basis for further research and application of this TCM-HP.
