Hybrid selection (5,7) and exon amplification (2,3) are two powerful methods to isolate genes from specific chromosome regions (positional cloning). In the process of positional cloning, several unknown genes may be isolated. Since sequence comparisonwith known genes in the GenBank~(R) database is the easiest way to get ideas about the possible function of these unknown genes, obtaining enough sequence information from these unknown genes is a major goal during the process of positional cloning. Unfortunately, most of the DNA fragments isolated by hybrid selection and exon amplification are small. Therefore, the sequence information from these unknown genes is limited.
