Endogenous tissue-type plasminogen activator (t-PA) is an endothelial serine protease that converts fibrin-bound plasminogen into plasmin that in turn lyses the fibrin. This property has been developed for the treatment of acute brain infarction (BI) by recornbinant t-PA (rt-PA). The activity of t-PA is controlled by plasminogen activator and inhibitor 1 (PAI-1). Tissue plasminogen activator antigen concentration primarily measures t-PA:PAI-l complex, which is determined by PAI-1 concentration. Hence, because PAI-1 was probably the determinant of the relationship observed between t-PA antigen concentration and arterial thrombotic disorders, increased t-PA concentration could reflect an impaired endogenous fibrinolysis .
