Rafflnose series oligosaccharides (RFOs) and phytic acid metabolism were studied in cotyledons, roots and shoots of chickpea seedlings at different days (1, 2, 3, 5 and 7) after germination (DAG). RFOs content in cotyledons depleted rapidly between 1and 2 DAG and completely by 3 DAG. Decreasing content of RFOs in cotyledons was correlated with combined activities of alpha-galactosidase and invertases, hence indicating the involvement of both enzymes in their hydrolysis. Depletion of RFOs and starchin cotyledons was accompanied by increase in sucrose content of cotyledons during germination. Hydrolysis of phytic acid in cotyledons during germination was correlated with combined activities of acid phytases and phosphatases. Very high activities of phosphatases were observed in cotyledons, roots and shoots during germination whereas activities of alkaline phosphatases were many folds lower than those of acid phosphatases. Phosphatase activities were positively correlated to water soluble inorganic phosphorus contents and negatively correlated to phospholipid-phosphorus contents in germinating cotyledon, roots and shoots. However water soluble organic phosphorus contents were not well correlated to phosphatase activities. On comparing the differenttissues, root and shoot had higher contents of sucrose, fructose, glucose and higher enzymes (invertases, alpha-galactosidase, acid phytase, acid phosphatase and alkaline phosphates) activities than those of cotyledons.
