High frequency plantlet regeneration via organogenesis in Solanum aculeatissimum Jacq. and possible exploitation of solasodine

We report here in vitro plant regeneration via organogenesis from leaf explants and production of solasodine from Solanum aculeatissimum. Best shoot regeneration was achieved from IAA-derived leaf callus when transferred to MS medium containing IAA-(0.5 mg/l) and kinetin (4 mg/l), which gave the maximum number of shoots (38 +- 0.6) per g callus. Micro shoots were rooted on MS medium containing IBA (1 mg/l). The regenerated plantlets were successfully established in soil with 90 percent survival. Solasodine content analysed in the callus cultures showed an increase concomitant to successive stages of shoot morphogenesis. Maximum solasodine yield was obtained from shoot-forming cultures incubated for 60 days in MS medium supplemented with IAA (0.5 mg/l) and kinetin (4 mg/l). Ten per cent of the regenerated plants showed a marginal increase in solasodine content over seed-derived plant.
