The effects of staining methods, number of spermatozoa examined, microscopic magnification and technician on the accuracy of assessment of ram sperm head morphology using computer-assisted sperm morphometry analysis were assessed. Frozen semen from10 fertile rams was thawed and prepared on slides for morphometric analysis. The best results were obtained when spermatozoa were stained with haematoxylin and Rose Bengal stains and at least 100 spermatozoa were measured at X40 magnification. With thisprocedure, a sample could be analysed in~3 min. No significant differences in sperm head measurements were detected between 2 technicians. Sperm head dimensions were as follows: length, 8.08μm; width, 4.80μm; width:length ratio, 0.59; area, 29.13μm2;perimeter, 23.93μm. The coefficient of variation for individual analyses ranged from 4.8% for length to 6.0% for area. The variation between replicates wasless than or =2.4%.
