Catechol-O-methyltransferase (COMT) has long been considered as a candidate for vulnerability to schizophrenia in view of both its function and location in the genome (Goldberg et al., 2003; Stefanis et al., 2004; Harrison and Weinberger, 2005). Data have been reported showing allelic association of the Vall58Met polymorphism of the COMTgene with the clinical manifestations and the response to or dose requirements ofantipsychotics in schizophrenia (Illi etal., 2003; Inada et al., 2003; Bertolino et al., 2004, 2007; Hagen et al., 2008), though negative associations of COMTgene with schizophrenia were found in Chinese Han and other populations (Daniels et al., 1996; Chen et al., 1999; Fan et al., 2002; Galderisi et al., 2005; Ho et al., 2005; Yu et al., 2007). Therefore, it is hypothesized that the functional polymorphism of the COMTgene could be related to drug response to antipsychotics in Chinese Han schizophrenia because of the role of COMT in dopamine catabolism.
