摘要

Probes that provide a continuous fluorescent readout of protein tyrosine kinase activity offer a direct means to observe kinase action in living cells,may serve in a diagnostic capacity as sensors of aberrant activity,and will undoubtedly prove invaluable in high-throughput screening assays.Several genetically encoded FRET-based proteins have been described that,upon tyrosine phospho-rylation,display fluorescent changes up to 50%.
