Assessment of adipogenic, antioxidant, and anti-inflammatory properties of whole and whey bovine colostrum

Bovine colostrum (BC) has been used for nutraceuticalpurposes for animals and humans. Bovinecolostrum is a complex heterogeneous product andits antimicrobial activity, antioxidant potential, andgrowth factors can vary depending on age and speciesof the cow as well as their environment. Bovine colostrumpreparation in skimmed or whey fractions canalso alter properties of BC. Our goal was to comparecumulative anti-inflammatory, antioxidant, and adipogenicproperties of natural (whole) versus whey BC. Wecompared properties of whole and whey BC in 3T3-L1preadipocytes permanently transfected with reportersresponding to changes in inflammatory (NfκbRE/greenfluorescent protein), anti-inflammatory (Nrf2/YFP),and adipogenic (Fabp4/cyan fluorescent protein) statusin cells. Interleukin-6 secretion in these cells wasmeasured by ELISA. Whole and whey BC induceIL-6 secretion from 3T3-L1 fibroblasts; however, wheypreparation stimulated less IL-6 secretion. Cumulativeinflammatory nuclear factor (NF)κB activation in thepresence of lipopolysaccharide was reduced by bothwhole (−27%) and whey BC (−22%) compared with lipopolysaccharide-treated cells (100%). Treatment withwhole BC was more effective in the reduction of NFκBactivation compared with whey BC and occurred in adose-dependent manner. In consonance with decreasedNFκB activation, the Nrf2 promoter activity wasalso reduced in response to whole (−27%) and whey(−13%) treatments compared with nontreated cells(100%). Whole and whey BC suppressed adipogenesis,measured as induction of Fabp4, by −27 and −13%,respectively, compared with nontreated 3T3-L1 fibroblasts(100%). Our results showed distinct differencesin properties of whey and whole BC that could be usedto attain reduced adipogenic or cumulative inflammatoryresponses.
