In drawing up guideline VDI 4254 Part 2, which describes the emission measurement of endotoxins at operational facilities, comparison measurements with Limulus Amoebocyte Lysate (LAL)- and recombinant Factor C (rFC)-tests were made and performance characteristics for the complete measurement procedure were determined according to guideline VDI 4219. For this purpose, 25 parallel samplings (double determinations) using impingers were carried out at a poultry house at high levels of bacterial emissions. The endotoxin activities were measured by five laboratories applying LAL- and rFC-tests from various manufacturers.The results showed that both methods in principle are suitable for the analysis of emission samples but yield different concentration levels. Due to the fact that rFC-tests do not need horse shoe crabs and show a lower measurement uncertainty compared to the LAL-test, the analysis of endotoxin activities in emission samples according guideline VDI 4254 Part 2 was described and standardized only for the rFC-test.
