Effects of serotonin re-uptake inhibition and amino acids on the neuroendocrine system and physical and cognitive fatigue were investigated. 10 male subjects cycled in 4 trials until exhaustion. Participants ingested a placebo in trial (T) Ⅰ, 20 mg paroxetine in T Ⅱ, 21 g branched-chain amino acids (BCAA) in T Ⅲ and 20 g tyrosine (TYR) in T Ⅳ. Plasma BCAA in T Ⅲ and plasma TYR in T Ⅳ were increased after 30 min of exercise. Heart rate and capillary lactate did not differ in trials. Plasma ammonia increments were higher in T Ⅲ. Supplementations did not affect plasma ACTH, cortisol and growth hormone. Prolac-tin increments were higher in T Ⅳ. Exhaustion was reached earlier in T Ⅱ. Drive during psychometric testing subsequent to exercise was improved in T Ⅲ and Ⅳ. The results indicate that fatigue during endurance exercise was influenced by pharmacological modification of the central serotonergic system. BCAA or TYR supplementation did not improve physical performance although mental performance might have been improved. Hormonal responses did not correlate with these changes in performance.
