Nkx2.5isexpressedinthecardiogenicmesodermofavian,mouse,andamphibianembryos.Tounderstandhowvariouscardiacfateswithinthisdomainareapportioned,wefatemappedthemesodermalXNkx2.5domainofneuraltubestageemXenopus/emembryos.ThelateralportionsoftheXNkx2.5expressiondomainintheneuraltubestageembryo(stage22)formthedorsalmesocardiumandroofofthepericardialcavitywhiletheinterveningventralregionclosestoformthemyocardialtube.XNkx2.5expressionismaintainedthroughouttheperiodofhearttubemorphogenesisanddifferentiationofmyocardial,mesocardial,andpericardialtissues.Aseriesofmicrosurgicalexperimentsshowedthatmyocardialdifferentiationinthelateralportionofthefieldissuppressedduringnormaldevelopmentbysignalsfromtheprospectivemyocardiumandbytissueslocatedmoredorsallyintheembryo,inparticulartheneuraltube.ThesesignalscombinetoblockmyogenesisdownstreamofXNkx2.5andatorabovethelevelofcontractileproteingeneexpression.WeproposethattheentireXNkx2.5/heartfieldistransientlyspecifiedascardiomyogenic.Suppressionofthisprogramredirectslateralcellstoadoptdorsalmesocardialanddorsalpericardialfatesandsubdividesthefieldintodistinctmyogenicandnonmyogeniccompartments./p/div
