Cellsperformwidevarietiesoffunctionsthatarefacilitated,inpart,byadoptinguniqueshapes.Manyofthegenesandpathwaysthatpromotecellfatespecificationhavebeenelucidated.However,relativelyfewtranscriptionfactorshavebeenidentifiedthatpromoteshapeacquisitionafterfatespecification.HereweshowthattheNkx5/HMXhomeodomainproteinMLS-2isrequiredforcellularelongationandshapemaintenanceoftwotubularepithelialcellsintheemC.elegans/emexcretorysystem,theductandporecells.TheNkx5/HMXfamilyishighlyconservedfromseaurchinstohumans,withknownrolesinneuronalandglialdevelopment.MLS-2isexpressedintheductandpore,anddefectsinemmls-2/emmutantsfirstarisewhentheductandporenormallyadoptuniqueshapes.MLS-2cooperateswiththeEGF-Ras-ERKpathwaytoturnontheLIN-48/Ovotranscriptionfactorintheductcellduringmorphogenesis.TheseresultsrevealanovelinteractionbetweentheNkx5/HMXfamilyandtheEGF-Raspathwayandimplicateatranscriptionfactor,MLS-2,asaregulatorofcellshape./p/div
