TheinvolvementofemSOX10/emandemZFHX1B/eminWaardenburgndash;Hirschsprungdisease(hypopigmentation,deafness,andabsenceofentericganglia)andMowatndash;Wilsonsyndrome(mentalretardation,facialdysmorphyandvariablecongenitalmalformationsincludingHirschsprungdisease)respectively,highlightedtheimportanceofbothtranscriptionfactorsduringentericnervoussystem(ENS)development.TheexpressionandfunctionofSOX10arenowwellestablished,butthoseofZFHX1Bremainelusive.HerewedescribetheexpressionprofileofemZfhx1b/emanditsgeneticinteractionswithemSox10/emduringmouseENSdevelopment.ThroughphenotypeanalysisofemSox10;Zfhx1b/emdoublemutants,weshowthatacoordinatedandbalancedinteractionbetweenthesetwogenesisrequiredfornormalENSdevelopment.DoublemutantspresentwithmoresevereENSdefectsduetodecreasedproliferationofentericprogenitorsandincreasedneuronaldifferentiationfromE11.5onwards.Thus,jointactivitybetweenthesetwotranscriptionfactorsiscrucialforproperENSdevelopmentandourresultscontributetotheunderstandingofthemolecularbasisofENSdefectsobservedbothinmutantmousemodelsandinpatientscarryingemSOX10/emandemZFHX1B/emmutations./p/div
