Trophoblastcellsarecharacterizedbyaninvasivebehaviorintothesurroundinguterinetissue.Inrodents,anearlyperi-/endovasculartypeofinvasionexertedbytrophoblastgiantcellscanbedistinguishedfromalateinterstitialtypecarriedoutbyglycogentrophoblastcells.Analysisofthemolecularmechanismsoftrophoblastinvasionhasbeenhampered,however,bythecomplextemporalandspatialpatternsofinvasion.Weutilizedtrophoblaststem(TS)celllinestostudytrophoblastinvasioninvitroandtoestablishamodelthatfacilitatesinvestigationofthisprocessonthemolecularlevel.OurresultsshowedthattrophoblastgiantcellsthatdifferentiatefromTScellsinvitroarecapableofpenetratingareconstitutedbasementmembranematrix.Consequently,invasionrateswereincreasedinvariousgiantcelldifferentiation-promotingconditions.WealsoderivedTScelllinesthatarehomozygousforamutationoftheemHand1/emtranscriptionfactor.TheemHand1/emsupminus;/minus;/supTScellsshowedreducedlevelsofgiantcelldifferentiationandexhibitedanapproximately50%decreaseininvasionrates.Insummary,trophoblastgiantcellsthatdifferentiatefromTScellsinvitrorecapitulatetheinvasivecapacityofnormaltrophoblastcellsinvivo.TheTScellsystemisavaluabletooltoidentifyandquantitativelystudyregulatorsoftrophoblastinvasion./p/div
