Self-incompatibilityinBrassicaoleraceaisnowviewedasacellularinteractionbetweenpollenandthepapillarcellsofthestigmasurface.Inthisspecies,theinhibitionofself-pollenoccursatthestigmasurfaceundertheinfluenceofS-locusspecificglycoproteins(SLSG).WeusedantibodiesspecificforaproteinepitopeofSLSGtostudythesubcellulardistributionofthesemoleculesinthestigmaticpapillae.TheantibodieshaveuncoveredaninterestingepitopepolymorphisminSLSGencodedbysubsetsofS-alleles,thusprovidinguswithusefulgeneticcontrolstodirectlyverifythespecificityoftheimmunolocalizationdata.ExaminationofthinsectionsofBrassicastigmasfollowingindirectimmunogoldlabelingshowedthatSLSGaccumulateinthepapillarcellwall,atthesitewhereinhibitionofself-pollentubedevelopmenthasbeenshowntooccur.Inaddition,theabsenceofgoldparticlesoverthepapillarcellwallsintheimmaturestigmasofveryyoungbuds,andtheintenselabelingofthesewallsinthestigmasofmaturebudsandopenflowers,correlateswellwiththeacquisitionoftheself-incompatibilityresponsebythedevelopingstigma.
