Developmentalregulation,fromthefetalperiodto11monthsofage,andtheinfluenceofdenervationontheappearanceanddisappearanceofthemolecularformsofacetylcholinesterase(AchE)andbutyrylcholinesterase(BuchE)inratskeletalmusclewereexamined.Theenzymeformswereextractedfromanteriortibialisin0.01Msodiumphosphatebuffer,pH7.0,containing1NNaCl,0.01MEGTA,1%TritonX-100,andacocktailofantiproteases,andanalyzedbyvelocitysedimentationon5a€“20%linearsucrosegradients.Threeprincipalforms,denotedbysedimentationcoefficientsof4,10.8,and16S,wereobservedinmusclefromallagegroups.TheamountsofeachofthemolecularformsofAchEandBuchEinskeletalmuscleexhibiteddistinctandreciprocalpatternsofappearanceanddisappearanceduringpre-andpostnataldevelopment.Intissuederivedfromanimalslessthan2weeksofage,BuchErepresentedthepredominantcomponentofactivityinthe4Sform,waspresentequallywithAchEinthe10.8Sform,andwassubordinatetoAchEinthe16Sform.Between1and2weeksofageaprogressiveincreaseinAchEactivitiescoincidentwithareductioninBuchEactivitiesresultedininversionintheamountsofthetwoenzymespresentinadultmuscle.DenervationofmusclecausedadramaticreductioninthepresenceofAchEmolecularformswithnodiscernableinfluenceonthepresenceofBuchEmolecularforms.TheseresultsindicatethatbiosynthesisofBuchEisstrictlyregulatedinareciprocalmannerwiththatofAchE,andthatBuchEmetabolismisindependentofthestateofmuscleinnervation.IncreasedsynthesisofAchEandeitherreducedsynthesisorincreaseddegradationofBuchEcanaccountforthereciprocalregulationoftheseenzymes.Thesecharacteristicsofmammalianmusclecontrastsharplywithcharacteristicsdeducedforaviantissue(Silmanetal.(1979)Nature(London280,160a€“162).Theinnervation-independentmetabolismofBuchEandthediversemodesofitsregulationindifferenttissuefromdifferentspeciessignifythatBuchEfunctionmaybeunrelatedtocholinergicneurotransmission.
