Cellsfromnewbornrathindlimbshowgreatlyenhancedmyogenicitywhengrownonsurfacescoatedwithpoly-l-lysinefollowedbylaminin(PLL/Lam)insteadofthecollagensroutinelyused.Coatingwithpoly-l-lysine(PLL)aloneorwithPLLfollowedbycollagendoesnotenhancemyogenicity.Bothmyogenicandnonmyogeniccells,asdistinguishedbyamonoclonalantibodyspecificfordesmin,attachequallywelltocollagen-andlaminin-coatedsurfaces,butthereisatwo-tofive-foldincreaseinthenumberofmyogeniccellsonPLL/Lamby72hr,followedbyincreasedmyotubeformation.TodeterminewhetherthisincreaseinmyogeniccellswasaconsequenceofaselectiveincreaseinproliferationonPLL/Lam,incorporationof5-bromodeoxyuridineintoDNAfollowedbylabelingwithanti-BrdUrdantibodywasusedasanindexofcellproliferation.Theresultsindicatethatdesminisexpressedinreplicatingratmyoblasts,andthatreplicationofmyogeniccellsisgreatlyenhancedonlaminincomparedtocollagen.Therateofreplicationofnonmyogeniccellsisthesameonbothsubstrates.Additionof10??g/mllaminintothemediumofcellsseededonPLLorcollagenhasnoeffectonmyogenicity.Weconcludethatalamininsubstrateenhancesskeletalmyogenesisinvitrobypromotingselectivelythereplicationofmyoblasts.Culturespreparedfromfetusesat17and19daysgestationalsoshowenhancedmyogenicitywhengrownonPLL/Lam,whilethosefrom15-dayfetusesdonot.Growthanddevelopmentoffetalmyoblastsoncollagenwereverypoor,whereasmyoblastsfromthenewbornratdoproliferateanddifferentiateonthissubstrate.Thusmyogeniccellsatdifferentstagesoffetalandneonataldevelopmentmayrequireandrespondtodifferentextracellularenvironments.MyotubeformationintheE63cloneofL8ratmyoblastsisinhibitedbyPLL/Lam.
