Excessivereactiveoxygenspecies(ROS)playakeyroleinthepathogenesisofdiabeticnephropathy.Thethioredoxin(TRX)system,amajorthiolantioxidantsystem,regulatesthereductionofintracellularROS.Hereweshowthathighglucose(HG)inhibitsTRXROS‐scavengingfunctionthroughp38mitogen‐activatedproteinkinase(MAPK)‐mediatedinductionofthioredoxininteractingprotein(TXNIP)inmousemesangialcells(MMCs).KnockdownofTXNIPinMMCsreversedHG‐inducedreductionofTRXactivityandinhibitedHG‐inducedactivationofp38MAPKandincreasedsynthesisofTGF‐β1andfibronectin.ThesedatasuggestthatHG‐inducedoverexpressionofTXNIPinMMCs,whichmaybeviathep38MAPKpathway...
