Weproposethattheproperevolvingunitinenzymeevolutionisnotasingle“fittestmolecule”,butaclusterofrelatedvariantsdenoteda“quasi‐species”.Adistributionofvariantsprovidesgeneticvariabilityandtherebyreducestheriskofinbreedingandevolutionarydead‐ends.Differentmatricesofsubstratesoractivitymodulatorswillleadtodifferentselectioncriteriaanddivergentevolutionarytrajectories.Weprovideexamplesfromourdirectedevolutionofglutathionetransferasesillustratingtheinterplaybetweenlibrariesofenzymevariantsandligandmatricesintheidentificationofquasi‐species.Theligandmatrixisshowntobecrucialtotheoutcomeofthesearchfornovelactivities.Inthissensetheexperimentalsystemresemblesthebiologicalenvironmentingoverningtheevolutionofenzymes...
