Wehavereportedthatvascularendothelialgrowthfactor(VEGF)‐Ainducestheproliferationofhumanpulmonaryvalveendothelialcells(HPVECs)throughnuclearfactorinactivatedTcells(NFAT)c1activation[1].HereweshowthatVEGF‐AincreasesthemigrationofHPVECsthroughNFATc1activation,suggestingthatVEGF‐A/NFATc1regulatesthemigrationofHPVECs.Tolearnhowthispathwaymaybeinvolvedinpost‐natalvalvularrepair,HPVECsweretreatedwithVEGF‐A,withorwithoutcyclosporineAtoselectivelyblockVEGF–NFATc1signaling.DownSyndromecriticalregion1(DSCR1)andheparin‐bindingEGF‐likegrowthfactor(HB‐EGF)aretwogenesidentifiedbyDNAmicroarrayasbeingup‐regulatedbyVEGF‐Ainacyclosporine‐A‐sensitivemanner.DSCR1silencingincreasedthemigrationofovinevalveendothelialcells,whereasHB‐EGFsilencinginhibitedmigration.ThisdifferentialeffectsuggeststhatVEGF‐A/NFATc1signalingmightbeacrucialcoordinatorofendothelialcellmigrationinpost‐natalvalves...
